Sermorelin is a synthetic 29-amino-acid peptide corresponding to the first 29 amino acids of natural GHRH. It was FDA-approved in 1997 for diagnostic testing of GH deficiency and has the longest clinical track record of any GH-releasing peptide.
Sermorelin has a shorter half-life (~10 minutes vs CJC-1295's 30 minutes without DAC), requiring more frequent dosing. However, its shorter action more closely mimics natural GHRH pulsatility and gives more precise control over GH stimulation. Many clinicians prefer sermorelin for its predictable, well-characterized pharmacology.
Unlike exogenous GH, sermorelin works through the pituitary's natural feedback mechanisms. If GH levels are already adequate, the pituitary reduces its response — making it nearly impossible to reach dangerous GH levels. This built-in safety mechanism is a major advantage.
Sermorelin binds to GHRH receptors on somatotroph cells in the anterior pituitary, stimulating cAMP-mediated synthesis and pulsatile release of endogenous GH. The response is modulated by somatostatin feedback, providing a self-limiting safety mechanism that prevents GH excess.
